DEMOMethod
A guinea pig is killed by a blow to the head and cutting the throat. The abdomen is opened and the ileum located. A short section (1-2 cm) of the ileum is removed and placed in a dish containing Tyrode's solution. It is important to avoid handling the tissue too much particularly with metal instruments and to avoid stretching the tissue. It is then tied onto a piece of glass tubing containing a metal electrode such that the exposed portion of the electrode protrudes into the lumen. The upper end of the intestine is attached to an isotonic transducer. Deflection of the pen is proportional to the amplitude of the contraction and in this experiment that is displayed as R (arbitrary units). The lumen of the gut should not be closed off at the top. A second electrode is dipped into the bathing solution and both electrodes are attached to an isolated stimulator allowing transmural stimulation of the preparation.
